You need to get the devon scope (from inside the devon corp after completing the main storyline. Then you can go to the beryl ward, and try to enter one of the caves in rhodocrine jungle. You will find an invisible object in your way, then go to silph co.

I mainly used my Steel and Poison types to fight him so for me he was generally easier than most other leaders, so you might want to teach one of your pokemon a TM or something.

Yes the chapter ends after you defeat him.

If you haven't already I think you have to battle the other gang leader somewhere in 7th street (can't remember exactly where he is), then your gang leader will appear somewhere at the top, at least that's what I had to do before he would appear. Speak to him and he will tell you about wanting to disband the team and so on, then you're on your way collecting applications.
